Triciribine is a small molecule DNA synthesis inhibitor. It can inhibit Akt and HIV-1 with IC50 values of 130nM and 20nM, respectively; however, it has no inhibitory effect on PI3K/PDK1; and its activity decreases by 5000 times in cells lacking adenosine kinase. At low concentrations, Triciribine exhibits antiviral and antitumor activity. Triciribine can effectively inhibit tumors in mice with high Akt expression. It reduces the survival of melanoma cells with high Akt phosphorylation and enhances the tumor growth inhibition mediated by ad-PUMA. Additionally, when used in combination with ad-PUMA,Triciribine significantly inhibits the growth of human melanoma in vivo. Triciribine has already been used in a phase II clinical trial for melanoma patients. Recent research in the U.S. has found that the anticancer drug Triciribine can delay or reverse lethal pulmonary fibrosis and pulmonary hypertension.
